Abstract

Cancer is one of the genetic disease that rated as the prime cause of death
and still rise every years. The current therapy for cancer are chemotherapy,
radiation, surgery, and gene therapy, but still have some weaknesess. One of the
therapy that still be developed is Photodynamic Therapy (PDT) that has higher
selectivity to damage cancer tissue since it exposed by photosensitizer, light and
oxygen at once become cytotoxic agent’s target during PDT. Zinc Pthalocyanine
(ZnPc) as photosensitizer compound from porphyrine derivate that has tetrapyrol
macrocyclic structure. The aim of this research is to know the physicochemical
characters of ZnPc for determine the potential use in PDT. From the result of
absorption prediction showed the lipophilicity that facilitated membrane
absorbtion but cannot accumulated. Then the result of absorption spectrum and
molar absorptivity there were 2 peaks, λ 605 nm (Q2) and λ 669,5 nm (Q1). The
Q1 band was effectively used for photosensitizer excitation at a lower
concentration and effectively used for cancer therapy in deeper tissues. The result
of ΦΔ test, 1O2 were formed higher at pH 6,3 than at pH 7,4 so that ZnPc was
more effective to damage cancer cells instead normal cells. In other hand, the
result of ΦF test was 0,3643 so that ZnPc was more effective to fluorescence than
to produce 1O2. Then from stability test in indirect sunlight showed the
photodegradation of ZnPc although not significant. In conclusion that ZnPc
potentially used in PDT.
