Abstract
Breast cancer is one type of cancer with the highest mortality rate in the world.
The use of 5-FU in cancer therapy is less effective and has high side effects if
used with increased doses. Actinobacteria DHE 6-7 isolated is known to have a
similarity with Streptomyces indiaensis (99.67% homology) which has secondary
metabolites as anticancer drugs. Ethyl acetate extract of actinobacteria DHE 6-7
isolate (EEIAI DHE 6-7) has an Rf value that is equal to actinomycin D of 0.23.
EEIAI DHE 6-7 produces actinomycin D which can be used as an anticancer.
Cytotoxic test of EEAIA DHE 6-7 on T47D breast cancer cells had IC50 values
0.711 μg / mL and 5-FU had IC50 203.9 μg / mL. The combination of EEAIA
DHE 6-7 and 5-FU showed a synergistic value with an optimal CI value of 0.197
which was at a concentration of combination of EEAIA DHE 6-7 0.4 μg / mL and
31 μg / mL. The T47D Cell proliferation test reduced the decrease in cell counts at
0 until 72 hours, a combination of EEAIA DHE 6-7 and 5-FU and gave better
results compared to single concentration. EEAIA DHE 6-7 can be used as a
combination chemotherapy agent and can increase the sensitivity of T47D cells to
5-FU.
