Abstract

Background: Traditionally, people used leaves and stem of Colocasia esculenta
as wound healing plant and piper betle leaves as natural antiseptic in wound.
Leaves of Colocasia esculenta contains saponin, terpene, tannin, flavonoid,
anthraquinone, cardiac glycoside and alkaloid. Piper betle leaves contains
variety of essential oil i.e; hydroxy chavicol, chavibetol, estragiol, eugenol and
terpene.
Objective: The aims of this research was to determine the state of wound healing
in rabits which was given various combination of Colocasia esculenta leaves and
stem ethanolic extract, and also leaves of piper betle ethanolic extract and also to
prove any differences of wound healing activity.
Methods: In vivo test was done with observation of the state of wound in each
combination in rabits. This research used 7 groups of treatment, that were
combination of leaves and stem of Colocasia esculenta extract: Piper leaves
extract (75%:25%), (50%:50%), (25%:75%), Colocasia esculenta extract 100%,
Piper betle extract 100%, positive control (Betadine®) and solvent control. One
way analysis of variance used dummy variabel was done to determine the
differences of wound healing effectivity in each treatment.
Result: The result showed that combination of leaves and stem Colocasia
esculenta extract : Piper betle leaves extract (75%:25%) was the highest state of
wound. Result of anova dummy variable showed that F calculate = 5,588 (F
calculate> F table 2,37) and P=0,029 (P<0,05). Meaning that there were
significant differences of wound healing state in each treatment. Analysis of
secondary metabolic coumpound’s showed that Colocasia esculenta leaves and
stem ethanolic extract, and also leaves of Piper betle ethanolic extract contain
tannin.
Conclution: The higher concentration of leaves and stem of Colocasia esculenta
extract concentration, the higher wound healing state. Colocasia esculenta leaves
and stem ethanolic extract, and also leaves of Piper betle L. ethanolic extract
contain tanin compound
