Abstract

Dental plaque is dominated by Streptococcus mutans bacteria. Ethanol extract of
green betel leaf (Piper betle Linn) containing kavikol and gambir (Uncaria gambir
(Hunter) Roxb) containing catechins is known to have antibacterial power against
Streptococcus mutans bacteria, it can be developed into herbal toothpaste
formulations to prevent dental plaque from arising. This study aims to make herbal
toothpaste preparations of green betel leaf and gambier by maceration method and
find out the optimum herbal toothpaste formula that can inhibit the activity of
Streptococcus mutans bacteria with the well diffusion method. The treatments were
divided into 5 groups, namely 3% betel leaf extract, 3% gambir extract, extract
combination of 1.5% each, positive control and negative control. The results of the
analysis on the viscosity test and foam height measurements using the Friedman Test
non parametric test and the Wilcoxon advanced test (p> 0.05) showed that the
formulations affected the value of foam viscosity and height but storage time only
affected the viscosity value. While the results of the analysis of the antibacterial
activity of toothpaste using the non parametric Kruskal-wallis test (p <0.05) which
means that there are differences in the inhibitory zone values between the
formulations, then repeated Mann-Whitney follow-up tests were carried out. So that
it can be concluded that the paste teeth containing ethanol extract of green betel leaf
and gambier have the highest antibacterial activity against the growth of
Streptococcus mutans bacteria in FIII which is a combination of green betel leaf
extract and gambir of [16.78±1.6mm] greater than Pepsodent's brand of toothpaste
namely [12.26±1.5mm].
