Abstract
Collagen has a role in the body, but the collagen that is widely available
on the market generally comes from the skin and bone tissue of cows or pigs, the
safety and halal of which need to be taken into account, so an alternative source of
collagen is needed that is safe, halal and cheap, so the type of fish used is tuna.
This research aims to determine the differences in optimum extraction methods
with concentrations of 0.1 M and 0.05 M NaOH, 0.1 M, 0.5 M and 0.75 M Acetic
Acid, and 0.9 M and 1 M NaCl. .8 M. The parameters observed were pH test,
water content, ash content, FTIR functional groups and analysis using PCA
chemometrics. The results of extraction on skin and bones using the extraction
method at a concentration of 0.1 M NaOH, 0.75 M Acetic Acid, and 1.8 M NaCl
in 3 hours produced the highest yields, namely 3.634% and 1.740%, pH 7.26 and
7. .43 with SNI (6.0 -7.0), water content 5.48% and 8.93% SNI (maximum 14%),
and ash content 1.2% and 2.3% SNI (maximum 1%) . The extraction results using
various methods on sets of 11 skins and bones showed better results compared to
other methods, but for ash content all methods did not comply with SNI.
Meanwhile, the FTIR results show that the collagen functional groups are Amides
A, B, I, II, and III. Then, from the data results using PCA chemometrics, skin set
7, bone set 8 and bone set 9 are similar to standard collagen. So from these results
the optimal extraction method is the extraction method for skin set 7, bone set 8
and bone set 9.
