Abstract

Kencur (Kaempferia galanga L.) is a tropical plant that grows in many
regions in Indonesia and is used as an herbal plant for various treatments. This
study aims to determine the effect of auxin types on callus growth and growth in
vitro, determine the effect of BAP addition on callus growth and growth, and
determine the interactions between auxin types combined with BAP cytokinins.
The research design used was a Completely Randomized Design (CRD). The first
factor is the growth regulator auxin 2,4-D IBA and NAA, each concentrations of 1
ppm, 2 ppm, and 3 ppm. The second factor is the growth regulator cytokinin BAP
with concentrations of 0 ppm and 0.1 ppm. The observed variables are callus
morphology, callus color, fresh callus weight, callus dry weight, and qualitative
secondary metabolite test. The results showed that the treatment of cytokinin,
auxin and interaction (SXA) types did not significantly affect the fresh weight of
callus and dry weight of callus. It is suspected that the concentration used has not
been able to increase callus growth and propagation.
