Abstract
Binahong (Anredera cordifolia L.) is one of the plants that can be used to treat various types of diseases, because it contains saponins, alkaloids, polyphenols, anthocyanin, flavonoids, oleanolic acids, proteins, vitamin C. Cultivation of Binahong has begun to be implemented by tissue culture technique. The initial stage of tissue culture which had an important role in the success of the culture is explant sterilization. The most important thing in explant sterilization is to combine the efforts to get a sterile explant and keep the explant tissue from being damaged due to high disinfectant concentrations. The Old Efficiency Study of Chlorine Soaking (Ca[ClO]2) on Explants to Increase the Success of In Vitro Binahong Culture (Anredera cordifolia L.) has been carried out at the Agrotechnology Laboratory and Plant Engineering Laboratory, The Efficiency of Soaking Duration of Chlorin (Ca[ClO]2) in Various Ekplants to Increase The Success of Binahong (Anredera cordifolia L.) has been carried out at the Agrotechnology Laboratory and Plant Engineering Laboratory, Muhammadiyah University, Purwokerto from January 2019 - May 2019. The research design uses factorial completely randomized design (CRD) with two factors, namely the length of chlorine immersion and the type of explant. Each factor consists of 2 levels so that 4 treatment combinations are obtained and each treatment is repeated 6 times. Observation variables included: time of contamination, percentage of contamination, percentage of type of contaminant, callus induction time, percentage of callus induction, and percentage of live explants. The results show that the duration of soaking period of chlorine for 4 minutes was more efficient in increasing the success of tissue culture in explants of Binahong leaves, and the duration of soaking of chlorine for 10 minutes was more efficient in increasing the success of tissue culture in explants of Binahong buds. Leaf explants have a higher percentage of live explants at 86%, while explant of buds have a percentage of live explants of only 55.5%.
