Abstract
Background: In 2019, Indonesia was ranked 7th as the highest country with 19.5 million people with diabetes mellitus with most type 2. Most people use traditional medicine to support conventional therapy. Leunca plant is one of the plants that has antihyperglycemia properties. This study aimed to determine the total flavonoid and total phenolic levels of fresh leunca fruit ethanol extract, determine the antihyperglycemia activity of leunca fruit ethanol extract against the reduction in blood glucose levels in male wistar rats of alloxan-induced. Method: This study is non-experimental and experimental, measuring total phenolic and total flavonoid levels using UV-Vis spectrophotometry and using male rats wistar strain which are grouped into 6 groups, namely normal control (without treatment), negative control (Na CMC 0.5%), positive control (glibenklami 0.45 mg/kg rat body weight), group 1 (extract 400 mg/kg rat body weight), group 2 (ethanol extract 800 mg/kg rat body weight), and group 3 (extract 1200 mg/kg rat body weight) Results: Total flavonoid levels of leunca fruit ethanol extract obtained 261.817 mgQE/100gram and total phenolic content of leunca fruit ethanol extract which was 464.954 mgGAE/100gram. Statistical analysis of average relative blood glucose levels (%) in abnormal normality tests and non-parameteric tests showed differences between groups. Conclusion: Leunca fruit ethanol extract in this study had total flavonoid levels and total phenolic levels of 261.817 mgQE/100gram and 464.954 mgGAE/100gram. Leunca fruit ethanol extract proved the existence of antihyperglycemia activity and there were differences in antihyperglycemia activity between treatment groups.
