Rossele petals (Hibiscus sabdarifa L) is one of many nutritious plants used as anti-bacterial, anti-thrush, anti-diabetic, cough reliever , pain reliever, hypertension reduction. This study was aimed to optimize and determine the matrix of the optimal Lactose and Mannitol to get good lozenges through Simplex Lattice Design (SLD). The tablet made by using wet granulation method. The compositions used were lactose, mannitol, gelatin, magnesium stearate and aerosil. Tests carried out on the physical properties of tablets, weight uniformity, hardness of tablets and tablet fragility. Time disintegration and taste response. The data were analysed by using software Design Expert Trial ver 12.0.1.0., so that it could be known the effects of adding both compositions. The result of the study showed that the combinations of lactose and mannitol could dissolve or disintegrate in accordance with the specified time limit and the taste response test using the Likert scale given to respondents showed that it was acceptable by them. The optimum formula composition obtained by using the Design Expert software Simplex Lattice Design (SLD) was Lactose 114,211 mg and Mannitol 89,788 mg showed desirability 0,49.
