Abstract

The study aimed to determine the effect of variation in Pb concentration on colony diameter and indextoleranceof indigenous non-symbiotic fungi from Pb heavy metal contaminated land and Minimum Inhibitor Concentration (MIC) value of indigenous non-symbiotic fungi isolates from heavy metal contaminated shallots. Pb. The research was conducted in the Microbiology Laboratory, Biology Study Program, Teacher Training and Education Faculty, Muhammadiyah University of Purwokerto on July-August 2018. The research was arranged by using Factorial Completely Randomized Design (CRD) with two factors. The first factor was the concentration of Pb heavy metals, namely: L0 (Pb 0 ppm), L1 (25 ppm), L2 (50 ppm), L3 (100 ppm), and L4 (200 ppm). The second factor was fungi isolates, namely: I1 (Penicillium cf. oxalicum), I2 (Trichoderma cf. harzianum), I3 (Aspergillus section Nigri), I4 (Penicillium cf. dangerdii,) I5 (Rhizoctonia), I6 (Aspergillus section Nigri), I8 (Penicillium cf. citrinum), I9 (Penicillium sp.), and I10 (Aspergillus terreus Thom.). The results of the research were analyzed by using the F test and if it has significantly affected,it continued by using DMRT test at the 5% level. The resultsshowed the treatment of fungi (I) isolates had a significant effect in the diameter variable with the best isolate, Trichoderma cf. harzianum (8 cm). The treatment of Pb heavy metal concentration and the interaction of the treatment of Pb heavy metal concentration and type of isolate on the diameter variable were not significantly affected. In the indextolerance variable, all treatments and interactions have no significant effect. The Minimum Inhibitor Concentration (MIC) value could not be determined.
