Abstract

Nata is a layer of extracellular polysaccharides (cellulose) that is formed
by the capsule-forming microbes. It is a fermented food that is created by
Acetobacter xylinum, when it is grown in a liquid media containing sugar such as
coconut water, it will produce a white layer of nata that floated on the surface
media. The objective of this study is firstly to screen the variables that influence
such as the amount of the carbon source using sugar (C), amount of the nitrogen
source using urea, (N), amount of phosphate source using K2HPO4, (P), amount
of the source of vitamins using vitamin B syrup,(V), fermentation time (LF), pH
and the starter volume (JS) of nata de coco using Placket-Burman Screening
Design, secondly optimalize the three most influential variables in the
fermentation using the Response Surface Method. The result of Placket-Burman
Screening Design show that the fermentation time (LF) and the amount of carbon
source (C) are main factor that influence to yield, whereas the main interaction
variable are C, pH, and LF. Meanwhile the result of Central Composite Design
(CCD) show that the optimum yield for the amount of the carbon source using
sugar at 17.5 gr/500 ml of coconut water, pH 5, and fermentation time for 10
days, it is obtained yield at 90.365% and the organoleptis test result show that
respondents like taste and texture of it’s nata.
