Abstract

Cancer is the fifth leading cause of death in Indonesia. Cancer therapy done in three ways, ie surgical therapy, however its effective for primary cancer that normal is localized, xray therapy (radiation) can damage the normal tissue, especially the cell with rapid proliferation, while chemotherapy often induces a second primary cancer as a result of the carcinogenic properties owned by the compound used for chemotherapy. One cancer therapies are being developed photodynamic therapy (photodynamic therapy or PDT). PDT is a treatment that requires visible light, a photosensitizer and oxygen. PDT produces singlet oxygen as a species capable of destroying tumor. The model compounds used photosensitizer is tetrapirol class macrocyclic compounds, compounds MTPP (meso-tetraphenylporphine) and MTPC (meso-tetraphenylchlorin) is used as a model compound representing the macrocyclic tetrapirol pyrrole ring is not reduced and reduced. From the results predicted partition coefficient log P values obtained MTPP = 10.3 while MTPC = 10.18, so MTPP lipofilisitas properties > MTPC based on the value of log P of the two compounds, then there is the effect of the reduction of pyrrole ring. Then the results of measurements of absorption spectra and molar absorptivity (ε) of the compound MTPP and MTPC, in Q1 band MTPC are two bands are on Q1a and Q1b, tetrapirol macrocyclic compounds are reduced more potential as a photosensitizer. Q1a band MTPC compound effectively used for photosensitizer excitation with a lower concentration, and Q1b band MTPC compounds can be used for cancer therapy in deeper tissues. From the result test of quantum yield of singlet oxygen (ΦΔ) MTPC compounds > MTPP compound either at pH 6.3 and 7.4 so that the power of singlet oxygen MTPC > MTPP. In addition to the stability test results are not direct sunlight MTPC compounds more volatile than the compound MTPP, then the color of the test solution becomes pale, oxygen is known to be one of the causes photodegradation and bleaching the colors.
