Abstract
Background: Indonesia as Muslims country suppose increase food quality which is appropriate within Islam’s .role in Quran surah An Nahl verse 115 which forbids pork. On government regulation number 69 ahun 1999 article 10 set inclusion halal labels on food products, but still found the case of mixing pork in food products. One of the food potential mixed with pork is beef cornet. The sample of the product analyzed by Real Time PCR taken from traditional market and supermarket in Purwokerto. The method used is a Real-Time PCR using primers specific species ND5 mitochondrial DNA, the method has advantages compared with regular PCR, Real Time PCR has a faster processing time, accuracy and higher sensitivity Objective: the purpose of his research is to analyze pork contaminations on the beef cornet sold in Purwokerto through DNA amplification using Real Time PCR. Method: this non-exsperimental research is using Real Time PCR uses specific species primer of DNA mitochondria ND5. The method is better than usual PCR, it is faster, more accurate and more sensitive than usual PCR. Results: On qualitative result analyzing of electrophoresis agarose gel shows that DNA isolated by seen of the bands which has different thickness in each. While comparator sample which mix pork in concentration of 5%, 10%, 25% and 50% are able to be amplified. Sample based on the Real Time PCR amplification curve, there are no amplifications or pork contaminations on five samples of beef cornet sold in Purwokerto. Conclusion: There are no pork contaminations in all of beef cornets sold in Purwokerto.
