Lantana camara leaves has been studied having cytotoxic effect on A375 cells,
has antiproliferative activity against HEp-2 and NCI-H292 cells. This study aims
to observe the combination effect of ethanolic extract of Lantana camara leaves
(EEDT) and 5-Fluorouracil in cytotoxicity, antiproliferative, and the mechanism
of cell cycle inhibition. MTT assay is used in the cytotoxic test with incubation
time for 24 hours and modulation of cell cycle observed by Flowcytometry assay.
The result showed that EEDT have moderate potency of cytotoxicity (IC50
125μg/ml). The combination of EEDT and 5-FU at low concentration has an
antagonistic effect (CI>1,1), at high concentrations having a mild-moderate
synergistic effect (CI=0,7-0,9). The combination of EEDT and 5-FU was able to
provide proliferation effect of T47D cells at 24 to 72 hours compared with cells
control, but the antiproliferation effect of combination of EEDT and 5-FU was not
as good as a single 5-FU usage. The combination of EEDT and 5-FU increased
cell accumulation at sub-G1 and G1 phases.
