Recently, many traditional herbs contain chemical drug, one of which is prednisone in uric acid herbs. Prednisone is a corticosteroid medicines that effective as pain relievers. This study aims was to analyze prednisone in uric acid herbs by high performance liquid chromatography method, using reversed phase method with 243 nm UV detector, ODS (Octadecyl silica) C18 shimpack column (250mm x 4.6mm; 5μm ) as stationary phase, methanol and aquabidest (60 : 40) ⁄ as mobile phase. Isocratic elution with a flow rate of 1 ml/minute and injection volume 20 μl. The result of linearity test in the range 10-35 μg/ml gives r=0.9955 with the equation Y=25007.6X+ 136348. Prednisone retention time of 6.61 minutes with a limit of detection 2.96 μg/ml, limit of quantitation 9.85 μg/ml and percent of recovery 100.109% ± 0.821. Based on the results of research on 8 brands of uric acid herbs at Purwokerto, two herbs proven to contain prednisone each of 21.77 and 50.07 μg/mg.
