Abstract

Keloid is uncontrolled synthesis and excessive storing of fibroblast tissue
and collagen in skin surface. A rumput mutiara is one of potential herbs. It
contains many active substances like alkaloid, terpenoid, olenolat acid, steroid,
and saponin. Terpenoid content in the form of oleanolat acid has been proved to
reduce the proliferation of fibroblast cell by inhibiting G1 of the cell cycle. It
works by decreasing the expression of mRNA of the collagen I and III and
reducing TGF-β1, a type 1 of the collagen. This study is to test its ethanol extract
activity ovaer the NIH3T3 fibroblast cell. It is to determine and find out the cell
growth activity, the proliferation, and the inhibitance at the phase cycle of
NIH3T3 fibroblast cell after it is given the extract of the grass as the antikeloid.
The sitotoxic test was done by MTT method with EERM concentrations of 1000,
500, 250, 125 and 50 μg /mL. The antiproliferation test was done using doubling
time method in the EERM concentrations of 1/2 IC50, 1/4 IC50 and 1/8 IC50. The
observation of cell cycle was done using Flowcytometry method. The sitotoxic
test of EERM to the fibroblast cell results in IC50 value of 242 μg/ml. The
proliferation test indicates that the number cells of the EERM treatment is
significantly less that the control cell. This proves that EERM can inhibit the
speed of cell proliferation. The observation of cell cycle (EERM 30,25 and 121
μg/mL) shows an accumulation at S-Phase along with the addition of EERM
concentration.
