Abstract
Background: Estrogen is a steroid hormone that is produced mostly in women
and produced in small amounts in men. Some diseases associated with lack of
estrogen in the body can occur in women, especially in women who experience
post menopause.
Objective: This study aimed to determine the estrogenic activity of the leaves and
flowers ofJeruju (Acanthus ilicifolius) and EC50 of estrogenic activity were
determined using YES-assay.
Method:This study performed by in vitro using YES- assay. With series of
concentration of ethanol extract from leaf and flower of jeruju (Acanthus
ilicifolius) 50,100,200,400,800,1000,1200,1600,2000 μg/ml in DMSO.
Results: The ethanol extract of leaves of Jeruju (Acanthus ilicifolius) on YES -
hERα showed that the treatment by extract was proportional to amount of units β-
galactosidase. At a concentration of 50 ug / ml showed the lowest activity. And at
highest concentration 2000 ug / ml showed the highest β galactosidase enzyme
activity. EC50 leaf of ethanol extract from jeruju samples determined byYEShERα
was 28.960 μg / ml. While the results from ethanol extract of the leaves
YES - hERβ showed the lowest β-galactosidase activity in the smallest
concentration (50 ug / ml), and the largest activity at a concentration of 2000 ug /
ml. EC50 for ethanol extract of leaf samples determined by YES - hERβ was
29.292 ug / ml. In addition, the ethanol extract of flowers determined byYEShERα
exhibited the yelow colours from oNP. However, EC50 ethanol extract of
flowers could not be calculated based on the parameters of the four models of log
dose - response. While on YES - HERβ treated by ethanol extract of flowers
exhibited β-galactosidase enzyme activity at concentration of 100 ug / ml and the
highest concentration of 2000ug / ml. So the EC50 obtained for 1,809 x 108 ug /
ml.
Conclusion: The ethanol extract of leaves and flowers weeds exhibited very weak
estrogenic activity as determined by YES-hERα and YES-hERβ.
