Abstract
An increase in infectious diseases due increase in bacterial resistance to
antibiotics. Resistance can occur extrachromosomal or chromosomal. This study
aims to determine the cause of antibiotic resistance in bacteria multiresistant MG
46 and WK 45. MG 46 resistant to chloramphenicol and gentamicin and WK 45
resistant to amoxicillin and chloramphenicol. At isolates MG 46 and WK 45 did
not found plasmid, thus we analyze the chromosomal DNA. Chromosome analysis
was done by PCR using universal primer 16S rRNA and after that the results of
PCR restricted use the enzyme PstI to be cut sequences CTGCAG. Restriction
fragment of differences in results between the MG 46 and Bacillus cereus, that
there are two fragments of the MG 46 (∞ 700 bp and ∞ 900 bp) and the absence
of fragme nts on Bacillus cereus. The easiest possibility that replacement bases
adenine become cytosine at nucleotide number 7004 so that the sequence
CTGAAG become CTGCAG that can be recognized by PstI. And the results of
Streptococcus pneumoniae restriction can not be used to compare with the results
of restriction WK 45 because the result Streptococcus pneumoniae restriction
experienced star activity when compared with the NCBI database. Could be
concluded possibility of mutations in the 16S rRNA MG 46 that causes can be
recognized by the enzyme PstI.
