Abstract

Background: Tuberculosis is the deadliest infectious disease with 5000 deaths per
day. MDR-TB (Multidrug-resistant TB) is TB that is resistant to isoniazid and
rifampicin. Rhoeo spatacea is reported to have good antituberculosis activity. The
leaf extract of this plant has the potential to inhibit the growth of M.tuberculosis
from the original and MDR-TB strains. Methods: Screening the compounds in
Rhoeo spathacea through the PASS Online website. After that, molecular docking
was performed on compounds in Rhoeo spathacea with the wildtype and mutants
of katG using PyRx, then the results of the docking were compared with the binding
affinity values of the katG wild-type protein. The mooring results were then
visualized using Biovia Discovery Studio. Result: From the screening results of
Rhoeo spathacea compounds, it was found that four compounds had antimycobacterium
and antituberculosic activities with a Pa value of 0.5 – 0.7. From
the docking results it is known that the four compounds screened have good
interactions and affinity compared to Isoniazid. The visualization results show
similarities in amino acid residues. Conclusion: There are four compounds in clam
pineapple (Rhoeo spathacea) which are predicted to have moderate
antimycobacterial and antituberculosic activity. The four compounds have better
binding affinity values compared to Isoniazid. As a result of the docking, there are
similarities in the bonding of amino acid residues which indicates that the position
of the pineapple shell compound with isoniazid is in the same position.
