Abstract

Cultivation of kopyor coconuts using embryos from culture techniques can yield trees with 100% kopyor coconut production. However, the leaf sheath and fruit stalk easily detach from the tree, especially in the dwarf kopyor coconut type. This research aims to analyze the histological differences in the leaf sheath and fruit stalk between the deep-type kopyor coconut and the dwarf-type kopyor coconut. Two varieties of deep-type kopyor coconuts used are Banyumas Deep Kopyor (DB) and Banyumas Hybrid Kopyor (HB), while the dwarf-type kopyor coconuts used are Red Dwarf Cungap (KCM) and Green Sinumpur Dwarf Kopyor (GHS). Wet preparations of kopyor coconut leaf sheaths and fruit stalks were made by making cross-sections at the midpoint using a razor blade. The preparations were then stained with 1% fast green (30 seconds) followed by 0.5% safranin (1 minute). The preparations were observed using an Olympus BX 51 microscope (Olympus, Japan) at 40X magnification, and images were captured using an Olympus DP74 camera (Olympus, Japan). The obtained images were further analyzed using the ImageJ image analysis application to calculate the number of fiber vessel bundles and sclerenchyma (FVBS), the size of FVBS, and the total area of FVBS. The data obtained were analyzed using ANOVA at a 95% confidence level and followed by the Duncan Multiple Range Test (DMRT) at a confidence level. The research results show that in general, the histology of kopyor coconut leaf sheaths and fruit stalks is composed of upper epidermis tissue, hypodermis tissue consisting of collenchyma cells, sclerenchyma tissue, and ground tissue or parenchyma with scattered fiber vessel bundles (FVB) in the parenchymal tissue. The histology of the leaf sheath, especially in the middle part, shows the presence of sclerenchyma tissue between parenchymal tissue and fiber vessel bundles, while in the fruit stalk, sclerenchyma tissue is not found. Deep-type kopyor coconuts have leaf sheaths with a higher amount of sclerenchyma tissue compared to dwarf-type kopyor coconuts and have a wider total area of fiber vessel bundles and sclerenchyma (FVBS) (30 mm2 / 124 mm2) in the observed cross-sectional area compared to dwarf-type kopyor coconuts (22 mm2 / 124 mm2). The fruit stalk of deep-type kopyor coconuts has a larger size of fiber vessel bundles (1.1 mm2) compared to the dwarf type (0.7 mm2), although the total area of fiber vessel bundles does not differ among the four observed varieties of kopyor coconuts, approximately 26-29 mm2 / 124 mm2 cross-sectional area. The research results indicate that deep-type kopyor coconuts have a stronger histological structure in the leaf sheath and fruit stalk compared to dwarf-type kopyor coconuts
