Abstract

Background: Public warning published by the Food and Drug Supervisory
Agency (BPOM) Number PW.02.04.1.4.10.22.166 of 2022 states that 41
traditional medicines have been found containing chemical drug ingredients. One
of them is toothache herbal medicine containing chemical drug ingredients of
paracetamol. This study aims to identify and determine the levels of paracetamol
drug chemicals and obtain valid methods to be used in analyzing paracetamol
content in toothache herbal preparations circulating in Cilacap. Method: The
method used to analyze paracetamol content in 4 samples of toothache herbs
obtained directly from the herbal medicine depot in Cilacap is TLC-Densitometry,
using stationary phase silica gel F254 and mobile phase chloroform: ethanol (8:
1). Spotting detection is carried out by densitometry in a wavelength of 250 nm.
Results: The results of the method validation obtained are the value of the
correlation coefficient (r) = 0.996, LOD = 0.0005%, LOQ = 0.0017%. The
precision test is shown by the RSD value (1.127%), and the accuracy obtained is
98.817%. Conclusion: The validation results of the paracetamol analysis method
in toothache herbs using the TLC-Densitometry method have met the parameters
of linearity, LOD, LOQ, precision, and good accuracy, it can be said that this
method can be used to determine levels in samples of toothache herbs circulating
in Cilacap. Of the 4 herbal medicine samples, 3 samples (w, x, and y) were
positive for paracetamol with each level of 2.6% (w/w); 12.7% (w/w); and 10.9%
(w/w).
