Abstract
One of the obstacles to increase sugar production in Indonesia is the low
productivity of sugarcane plantations. Many efforts have been made to increase
sugarcane productivity in Indonesia, such as proper land preparation, correct
planting techniques, proper fertilization, pest and disease control, and the
provision of quality sugarcane. The way to produce quality sugarcane seeds is by
using meristem culture techniques, but the success of sugarcane meristem culture
is still not widely practised in Indonesia, especially at the acclimatization stage.
Therefore, this study aims to develop root induction techniques and
acclimatization of BL sugarcane varieties resulted from meristem culture.
Sugarcane plantlets of BL varieties resulted from meristem culture which did not
have roots were treated within Vitro on root induction medium for 28 days. The
root induction medium used was MS medium with the addition of myoinositol at
a concentration of 100 200 mg / l. Two types of medium used are liquid and solid
medium. The root-induced plantlets were then acclimatized using a mini growth
chamber, then continued with the second stage of acclimatization and treated in
moist conditions. The results show that root induction is successful with a high
success rate of 96-100%, both in liquid and solid medium with or without the
addition of myoinositol. The solid medium succeeds in increasing the number of
roots and the number of shoots, while the liquid medium actually decreases the
number of roots and the number of shoots. The plantlets are successfully
acclimatized with high success rates, 85-97% in the first stage and 82-88% in the
second stage. The resulting sugarcane seeds show normal and healthy growth
based on morphological characteristics of the roots number, root length, number
of shoots, shoot height, and weight on fresh. The results show that root induction
and acclimatization of BL sugarcane seeds resulted from meristem culture are
successfully carried out with high success rates and produce seeds ready for
planting into the land.
