Abstract

This study aimed to determine the best concentration of HgCl2 and immersion duration in the culture of galangal roots (Kaempferia galanga L) and to determine the effect of HgCl2 concentration and immersion time on the appearance of galangal root explants (Kaempferia galanga L). The design used was a Completely Randomized Design (CRD) consisted of two factors. The first factor was the concentration of HgCl2consisting of 3 levels of 0.1%, 0.3% and 0.5%. While the second factor was immersion time consisted of 3 levels of 1 minute, 3 minutes and 5 minutes. There were 9 treatment combinations repeated 6 times. The results indicated that the treatment of H0.3T3, H0.3T5, H0.5T3, H0.5T5 contamination rate of 0%. H0.1T1 treatment was 33.33% contamination, H0.1T3 treatment, H0.1T5, H0.3T1, H0.5T1 contamination rate was 16.66%. Duration of appeared contamination in the contaminated treatment varied from 4 to 22.5 DAP. Identification of the source of appeared contamination was suspected to be Aspergillus sp. Penicillium sp. and pseudomonas sp. The appearance of galangal root explants after explant sterilization did not experience loss or browning, visually the explant volume did not decrease, even though the explants did not experience signs of growth. The best treatment was H0,3T3 because the level of contamination was 0% with the use of a concentration of HgCl2 that was not too high. The appearance of explants of galangal root was white, no browning, visually the explants volume was not reduced
