Abstract

Conventional cultivation has not been able to meet the demand for galingale market nationally, propagation by callus culture techniques in vitro can be used as an alternative to provide galingale in large quantities. Propagation of tissue culture can be done in several ways, one of them is somatic embryogenesis. The aim of this research was to figure out the combination of NAA and BAP on the succeed of somatic embryo callus galingale. The research used a factorial complete design (FCD) with two factors of preparation and 6 replications. The first factor consists of 3 levels, which are the consentration of BAP 3 ppm (B3), BAP 4 ppm (B4), BAP 5 ppm (B5). The observed variables are each phase, the percentage of contamination and the percentage of callus growth. The results shows that using NAA did not involve real somatic embryogenesis of galingale callus while administration of BAP is real in each phase of embryogenesis, the interaction between NAA and BAP required a real initiative for growth, not in accordance with the reality in the liver and torpedo phases. The best treatment was the 5 ppm BAP (B5) treatment. The best interaction between NAA and BAP was in the treatment of B5N1 (BAP 5 ppm and NAA 0.25 ppm) in the shoot phase. Contamination rate is 0-11,1% with contamination sources of fungal.
