Abstract

One of the problems faced in tissue culture technique is medium contaminated by fungus. To control this, synthetic compounds of Plant Preservative Mixture (PPM) is the effort used. However, this is relatively expensiveand is usually hard to find. Therefore, natural alternative anti-fungus compound that comes from plants is needed. One of the potential plants is galangal (Alpinia galanga L.) due to its bio-active compound which has anti-fungus effect. The purpose of this research was to find out the effectiveness of galangal rhizome extract as the substitution of synthetic compounds role in hampering the growth of contaminant fungus isolates in MS tissue culture medium. This research used disc diffusion method by Kirby Bauer. The galangal rhizome extract tested was on the concentration of 15%, 20%, 25% and 30% with 3 times repetitions. Aquades (distilled water) was used as the negative control, while fungicide of Dithane M-45 1% as the positive control. The parameter observed wasthe width of hampered zone. The data were analyzed using F test and continued with Duncans Multiple RangeTest. The results showed there were 5 contaminant fungus isolates identified which were Aspergillus niger, A.tamarii, A. tubingensis, Gonatobotrys corda and Paecilomyces victoriae. Galangal rhizome extract was able to give hampering power toward the isolate of Aspergillus tubingensis fungus on all concentrations with the largest width on the concentration of 30% which was 375.45 mm2, and Paecilomyces victoriae fungus only on the concentration of 30% which was 187.73 mm2. However, the largest hampered zone from the treatment was so much smaller compared with the positive control which was 2007.74 mm2. Therefore, galangal rhizome extract is not effective yet to substitute the role of synthetic compound.
