Abstract

The prevalence of cancer in 2018 were 18.1 million new cases and 9.6 million deaths in the world based on 2018 GLOBOCAN data (IARC). One of the cancer treatments is 5-fluorouracil, but the effectiveness of 5-fluorouracil as a chemotherapy agent is still bad, so, a combination agent is needed to increase its effectiveness. Preclinical study showed that β-blockers had anticancer activity. One of a β-blockers group named atenolol, has been studied that it had the ability to reduce tumor size / metastasis. This study aims to determine the cytotoxic effect of the atenolol combination and 5-fluorouracil on T47D breast cancer cells, it is also to determine the effectiveness of atenolol combination and 5-fluorouracil on proliferation kinetics blocking for T47D breast cancer cells. The observation of cytotoxic effect was conducted by using the MTT assay method with 24 hours incubation time. The cytotoxic test of atenolol showed a potency of cytotoxicity with 285,51 μg / mL of IC50. The combination of atenolol and 5-Fluorouracil with various concentrations have strong synergistic effect (CI <0.3) until mildmoderate antagonistic effect (CI> 1). The number of T47D cells was rather decreasing at 48th to 72nd hour in the treated cells than the controlled cells, so the combination of atenolol and 5-Fluorouracil could increase the sensitivity of T47D breast cancer cells greater than the treatment of single atenolol tablet and 5- Fluorouracil for low dose.
