Abstract

Free radicalis atom or molecule that has unpaired electrons, very reactive and can causes oxidative stress. Oxidative stress in the body leads to several diseases such as cancer, atherosclerosis, coronary disease, and autoimmune diseases. Oxidative stress can be inhibited by substances called antioxidants. Mango (Mangifera indica L. ) leaves from Gedong varieties has a potential as a source of natural antioxidants. This study aims to isolate the antioxidant compounds present in the ethanol fraction of mango leaf (Gedong var.) using column chromatography and test its antioxidant activity using DPPH (2,2-diphenyl-1-picrylhydrazyl) assay. Extraction and fractination was carried out using n-hexane : ethyl acetate : ethanol. Ethanol fraction was then isolated by column chromatography with silica gel 60 as stationary phase and chloroform : acetone : formic acid as mobile phase. Qualitative analysis to test the presence of antioxidant compounds was carried out using thin layer chromatography, followed by DPPH spraying. The positive isolates from DPPH spraying were classified into isolate A, B, and C based on their Rf value. Further study was conducted on ethanol fraction, isolate A, isolate B, and isolate C with DPPH assay to determine the IC50 value. The result showed that IC50 value of ethanol fraction was 18.972 ppm, while isolate A, isolate B, and isolate C were 9.280 ppm, 27.753 ppm and 37.837 ppm, respectively, in comparison to vitamin C (8.010 ppm) as a positive control. Kruskal-wallis (p≤0.05) indicated a significant difference in each sample group. Mann Whitney test showed that isolate A has antioxidant ability that is comparable to vitamin C.
