Abstract
Background: Inflammation is a common symptom experienced by most people. The
choice of anti-inflammatory drugs is limited to use the NSAIDs. However the use of
these drugs risk of side effects in the gastrointestinal tract. Therefore, the use of
natural medicine or traditional medicine could been alternative for the treatment of
inflammation. One of the medicinal plants which are widely used for inflammation
treatment, is the Orthosiphon stamineus. However, there is no information available
for anti-inflammatory mechanism of Orthosiphon stamineus L.
Research Objectives: The aim of this study was to determine the antiinflamation and
antiagregation activity of ethanol extract of leaves of the orthosiphon stamineus L.
Results: In the present study used of six treatment groups in the Na-CMC (solvent
control), aspirin (positive control), Na diclofenac (positive control), the dose of 100
mg / kg, 500 mg / kg, and 2500 mg / kg body weight of ethanolic extract of
Orthosiphon stamineus L. In vivo test performed by the ear edema and rat paw edema
test. Data from both assays, then analyzed statistically by two-way anava followed by
the T-Test to compare the results of percent inhibition from those two it resulted that
doses of 500 and 2500 mg / kg did not differ significantly with the positive control.
From the T-test result, it showed that percent inhibition of inflammation shown in the
rat paw edema was greater than in the ear edema test. In vitro test performed by
platelet antiagregation which included the bleeding time and absorbance. Antiaggregation
test result data were analyzed with one-way anava. From the results of
statistical analysis of the bleeding time test and observation of the plasma absorbance
changes. From the result of statistical analysis from bleeding time test absorbance
changes of plasma using one-way anava, it exhibited that all doses of treatment had
no significant different with positive control.
Conclusion: Ethanol leaf extract of orthosiphon stamineus L. exhibited
antiinflammatory activity at doses of 500 mg/Kg and 2500 mg/kg BW as well as
showed antiagregation activity at doses of 100 mg / kg, 500 mg / Kg and 2500 mg /
kg body weight.
