Abstract

Background : Jeruju belongs to semi-mangrove plants which is considered as weeds by the people in coastal area. Several researches reported that extract of jeruju (Acanthus ilicifolius) exhibited antimicrobial activity, anticancer, antioxidant, hepatoprotective, antiinflamation, and hemolytic. One of secondary metabolite compound which exists in jeruju stem is lignan glucosides which is categorized as phytoestrogens. Thus, it is interested to observe about estrogenic activity in jeruju stem using YES-assay method. Aims of The Research : This research was aimed to determine estrogenic activity of water and ethyl acetate fraction of jeruju stem (Acanthus ilicifolius) and to determine its TLC profile. Research Methodology : Fractionation was done by using liquid-liquid fractionation. Estrogenic assay on water and ethyl acetate fraction of jeruju stem was done with concentration series of 50μl, 100μl, 200μl, 400μl, 800μl, 1200μl, 1600μl, 2000μl. The data was analyzed by using Graphad prism 4th version. Identification of fraction from extract of jeruju stem was done by using Thin Layer Chromatogram method (TLC). Result : Water fraction exhibited the highest estrogenic activity either determined by using YES-hERα or YES-hERβ, while ethyl acetate fraction showed the lowest estrogenic activity either determined by YES-hERα or YES-hERβ. The value of EC50 from water and ethyl acetate fraction determined by YES-hERα was 360,2 μg/ml and 4,369x1030 μg/ml. The value of EC50 from water and ethyl acetate fraction determined by YES-hERβ was 1083 μg/ml, and 1,34x106 μg/ml. From compound identification using TLC, it showed that water fraction contained flavonoid. While within ethyl acetate fraction contained flavonoid, alkaloid, saponin, tannin, and terpenoid. Conclusion : Water fraction exhibited low estrogenic activity either determined by using YES-hERα or YES-hERβ with negative relation between consentration and β-galaktosidase unit that was produced. Whereas ethyl acetate fraction of jeruju stem (Acanthus ilicifolius) didn’t have estrogenic activity either that determined by using YES-hERα or YES-hERβ. Compound identification used TLC to know the compound that was in water fraction of jeruju stem containing flavonoid, and within ethyl acetate fraction, it contained alkaloid, flavonoid, saponin, tannin, terpenoid.
