Abstract

Background: Metoprolol is an antihypertensive drug that has a high variability of each person when metabolized by CYP2D6 in the liver, consequently may increase the risk of adverse drug reaction and decrease in therapeutic response. As a rule over 95 % of an oral dose can be recovered in the urine which 10 % in unchanged form. Therefore to study the differences in drug metabolism among individuals can be determining the ratio of the levels of the parent drug and its metabolites in urine. Valid analytical method are needed to establish the levels of metoprololin the body, especially in the urine. Objective: This study aims to validate a method analize metoprolol in urine by high performance liquid chromatography. Method: High Performance Liquid Chromatography (HPLC) using reverse-phase LiChroCART®purospher® (250 mm × 3 mm i.d. × 5 µm) C18e (Merck) C18e column with mobile phase mixture of potassium dihydrogen phosphate pH 3, acetonitrile and methanol (85:10:5), flow rate 1.0 mL /min, and detected at a wavelength of 234 nm with a detector UV. The method was validated based on the parameters of accuracy, precision, linearity, LOD and LOQ. Results: The linearity result at 2-10 μg/mL showed r = 0.999, precision at concentrations of 4 and 8 μg/mL showed the value of RSD 0.014% and 1.215%, LOD and LOQ obtained value of 0.390 and 1.302 μg/mL, while the accuracy of the concentration of 4 and 8 μg/mL showed the value of % recovery 94.96 and 96.76 %. Conclusion: The results of this study concluded meet the validation requirements and can be used to determine the levels of metoprolol in urine.
