Abstract

Cacao (Theobroma cacao L.) is one of the main plantation commodities that has important role in Indonesian economic and Indonesia becomes the big second of producing the cacao in the world. However, the plantation production of cacao was low. One of the causes was the low of quality of the seed. Some effort had been done to the get high quality of the seed in large quantity through somatic embryogenesis technique, for example. Yet, the level of the success was categorized to be low. This research aimed to increase the success of the embryo induction of somatic through effectiveness test of the addition of 6-benzylamino purine (BAP) and vitamins with some different compositions to planting media toward the success of embryo induction of somatic cacao. The result of research showed that somatic embryo was successful to be inducted from friable callus bred in the medium of DKW (Driver & Kuniyuki, 1984) with the addition of 5 x 10-8 M BAP in 4 weeks with the success level of 2,2%. Friable callus was gained from staminodia explant and petals bred in the medium of DKW with the addition of 5 x 10-7 M kinetin and of 10-6 M 2,4-D. the addition of BAP to the planting medium with higher concentration (7 x 10-8 M) or lower (5 x 10-9 M and 10-8 M) was not successful to induct somatic embryo. The addition of vitamin with the various compositions (½ DKW, DKW, 1½ DKW, B5 (Gamborg et al., 1968) and MS (Murashige & Skoog, 1962) was actually proven not to affect on the weight increase of wet callus cacao and the success of somatic embryo induction, moreover it caused the callus morphology changes from friable callus to compact callus and it inducted the root of the planted callus. However, in the medium with BAP addition in low concentration (5 x 10-9 M), the composition of vitamin B5 affected significantly to increase the weight of wet callus cacao.
