Abstract

Mangroves produce secondary metabolites is estimated that there are 100,000-200,000 secondary metabolites and approximately 20% of the carbon. The compounds contained in Acanthus ilicifolius was saponins, flavonoids, polyphenols, compounds verbaskosida and phenolic acids (acid vanilat, siringat acid, ferulic acid, p-hydroxybenzoic acid and p-kumarat acid) which some of phytoestrogens. The content of these compounds allows interaction with the estrogen receptor so can be determine estrogenic activity. Objective : The purpose of this study was to determine the estrogenic activity of water fraction and ethyl acetate fraction of jeruju flower using YES assay methods that contain estrogen receptors alpha and beta and to determine the profile of the compounds found in the water fraction and ethyl acetate fraction of jeruju flower. Methods : The study was conducted using by Yeast Estrogen Screen (YES) Assay. The test is performed in vitro, the controls used 17-β-estradiol and g enistein were made with a concentration 10-4 M, 10-6 M, 10-8 M, 10-10 M, 10-12 M, 10-14 M, 10-16 M in DMSO and genistein with a concentration 10-5 M, 10-7 M, 10-9 M, 10-11 M, 10-13 M, 10-15 M, dan 10-17 M. Samples ethyl acetate fraction and water fraction flowers Acanthus ilicifolius is made with a concentration of 50 ug/ml, 100 ug/ml, 200 μg/ml, 400 ug/ml, 800 ug/mL, 1000 μg/mL, 1200 μg/mL, 1600 μg/mL and 2000μg/ml in DMSO. The screening compounds found in the water fraction and ethyl acetate fraction was carried out using thin layer chromatography (TLC). Results : The results estrogenic activity EC50 17-β-estradiol YES-hERα was no response, where as in the YES-hERβ is 2,0094x109M. EC50 of genistein YES-hERα is 7,8219x10-17M, where as in the YES-hERβ is no response. EC50 of estrogenic activity of ethyl acetate fraction of the YES-hERα was 335.9 μg/ml and the ethyl acetate fraction of the YES-hERβ was 1943 μg/ml. The water fraction of the YES-hERα was 764 μg/ml and the water fraction of the YES-hERβ was 0.0 μg/ml or no response. The compounds contained in the ethyl acetate fraction of jeruju include tannins, alkaloids, flavonoids, saponins and terpenoids while the water fraction contained the alkaloids, flavonoids and tannins. Conclusion :Ethyl acetate fraction flower jeruju has greater estrogenic activity than water fractions of jeruju flower determined by YES-hERα or YES-hERβ.
