Abstract

Estrogen is the hormone responsible for regulating cellular complex associated with differentiation, function and regulation of female reproductive tissues. Use of Hormone Replacement Therapy (HRT) can prevent these symptoms, but when used long term excessive risk of endometrial proliferation and endometrial cancer as possible. Phytoestrogens are natural decomposition of the plant that has many similarities with estradiol, a natural form of estrogen that is most potent. Acanthus ilicifolius plants contain compounds glucoside, alkaloids, flavonoids, fatty acids, steroids, lignans, and phenolic components and terpenoids. Based on these premises, then the weeds research estrogenic Acanthus ilicifolius. Objective: This study aimed to determine the fraction of ethylacetate and water fraction of leaves Acanthus ilicifolius have estrogenic activity were determined using the Yeast Estrogen Screen (YES) Assay. Methods: The study was conducted using by Yeast Estrogen Screen (YES) Assay. The test is performed in vitro, the controls used 17-β-estradiol and genistein were made with a concentration 10-4M, 10-6 M, 10-8M, 10-10M, 10-12M, 10-14M, 10-16 M in DMSO and genistein with a concentration 10-5M, 10-7 M, 10-9M, 10-11M, 10- 13M, 10-15M, dan 10-17 M Samples ethyl acetate fraction and water fraction leaf Acanthus ilicifolius L. is made with a concentration of 50ug/ml, 100ug/ml, 200μg/ml, 400ug/ml, 800ug/mL, 1000 μg/mL, 1200 μg/mL, 1600 μg/mL and 2000μg/ml in DMSO. Results: EC50 of 17-β-estradiol YES-hERα is no response, where as in the YEShERβ is 2,0094x109 M. EC50 of genistein YES-hERα is 7,8219x10-17 M, where as in the YES-hERβ is no response. EC50 in ethyl acetate fraction of the YEShERα is 28712 μg/ml , where as the YES-hERβ is 1327 μg/ml. EC50 in water fraction YES-hERα and YES-hERβ is no response. Conclusion: Ethyl acetate fraction of the leaf Acanthus ilicifolius L. had estrogen activity but lower than the fraction of leaf water weeds that do not have estrogen activity. The content of the compound in the ethyl acetate fraction include alkaloids, flavonoids, terpenoids, tannins and saponins, while the water fraction leaf weeds including flavonoids and tannins.
