Abstract

Background: Based on the data census, estimated number of women post-menopause in the world around 476 million in 1990. At least in 2030 this number will be increased to 1.200 million. One of the therapies that can been used was hormone replacement theraphy (HRT) i.e. estradiol. However estradiol had side effect such as breast cancer therefore the use of fitoestrogen as a substitute for intake of estrogen has been an option. Sunflower seeds contain several compounds that include fitoestrogen. But there were no scientific in report about the determination of estrogenic activity of sunflower seeds (Helianthus annuus L.) Aim: To determine the ability of daidzein replacing estradiol estrogen receptor binding and to determine estrogenic activity from extract ethanol of sunflower seeds (Helianthus annuus L.) using YES assay method. Method: This research was non-experimental in molecular docking and experimental in testing YES to determine estrogenic activity with series concentration of 50 μg/ml until 2000 μg/ml. Determining molecular docking by evaluate the value of RMSD and free energy. In YES test, increasing estrogenic activity marked by increasing yellow color from the formation of oNP. Finding: Free energy from ligan daidzein (sample) -2,60 kkal/mol and ligand estradiol (standard) -9,55 kkal/mol. Value of RMSD estradiol was 0,844 Ǻ. Value of EC50 sunflower seeds (Helianthus annuus L.) 981,2 μg/ml. Conclusion: Ethanol extract of sunflower seeds (Helianthus annuus L.) exhibited low estrogenic activity.
