Abstract

Background: The Extract of areca seed ethanol showed that the antioxidant activity IC50 of 0.409 mg / mL was greater than vitamin c as a positive control with IC50 value of 0.964 mg / mL. Based on these premises proves that areca seed extract has potential as an antioxidant yet there was no further research that isolate the chemical constituents of the various factions in the areca seed extracts. Therefore it was necessary to isolate chemical constituents of some fractions and antioxidant activity test of the extracts and various fractions of areca seed. Objective: The objective of this study was to isolate the chemical constituents of the fractions and tested the extracts of antioxidant activity and fractions of areca seed. Methods of the research: The study was carried out by radical DPPH for the methanol extract, fraction of methanol-water, the fraction of n-hexane, fraction of ethyl acetate, methanol fraction areca seed and to isolate the chemical constituents of the methanol fraction using Thin Layer Chromatography, preparative thin layer chromatography and spectrophotometry UV -Vis. Results: the result revealed that methanol fraction with IC50 is the smallest fraction (20.738 ppm) compared by the methanol-water fractions, fractions ethyl acetate and n-hexane fraction. It was because of the active compounds of areca nut adsorbed into methanol fraction. Therefore, those fractions have the greatest antioxidant activity. The method of isolation was using 84.6 mg TLC and TLC Preparative then it tested by the antioxidant activity with IC50 of 22396.3 ppm. Conclusion: The antioxidant activity of the methanol fraction was better than the methanol-water fractions, fractions ethyl acetate and n-hexane fraction. The Methanol fraction that had been isolated proved the active compounds as antioxidants.
